Spock's death, with the destruction of the old Enterprise coming a close second.

Spocks death is without a doubt the best of Trek. Followed closely by the send-off in TUC.

Honourable mention to Enterprise burning up in the atmosphere, Shatner did an excellent job with Kirk's emotion in that scene.

Its a hard call, Spock's death and farewell was exceptionally well done but in the end I have to go with the signing off in TUC, it marked the end of an era and I very nearly tear up when I see it.

I didnt really feel anything when the Enterprise died. Spocks death was great though.
What does defeat mean to you?

Nothing it will never come. Death before defeat. I don’t bend or break. I end, if I meet a foe capable of it. Victory is in forcing the opponent to back down. I do not. There is no defeat.

The most moving scene in Trek. I cried when I first saw it at a drive in theater in Texas, when I was around 7 years old( Dad! Spock died! Waaaahhh!!). Of course, the scene that most sticks in my mind from that time is when the worm crawls out of Chekov's ear.

Made me shed tears. Of course, then came the anger of "How could they do that!" Though, the death of the Enterprise was close.
No trees were killed in transmission of this message. However, some electrons were mildly inconvenienced.
